First, use alkaline dye crystal violet or gentian violet dye to stain the specimen, then add iodine solution to fix the dye on the stained object, use alcohol to decolorize some positive bacteria, and then transfer to the staining jar of fuchsin dye for a second staining.
All bacteria that are stained purple are called Gram-negative bacteria, and all bacteria that can be decolorized by alcohol and stained red are called Gram-positive bacteria.
Finally, rinse the excess dye on the glass slide with water, and observe the state of bacteria, flagella, and spores of bacteria through a microscope (because the flagella and spores of bacteria that have not been stained are transparent and it is not easy to observe their full appearance).
The staining method can be used to identify the type of bacteria.
